Retatrutide + MOTS-c
Quick Reference
Overview
This collection pairs one of the most extensively studied appetite-signalling peptides in current research with a mitochondrial peptide that research suggests influences how cells use energy. The two act at completely different scales — one on the brain’s appetite and blood-sugar signalling, the other on the tiny energy factories inside every cell.
Retatrutide has been among the most closely studied compounds in recent metabolic research, examined across Eli Lilly’s trial program for how it acts on the body’s metabolic signalling. MOTS-c, meanwhile, has drawn interest as a marker of mitochondrial health, with levels that shift alongside metabolic state. Researchers pair the two to study whether acting on intake signalling and cellular energy use together behaves differently from either alone.
The Components
Metabolic Peptides
Retatrutide
A next-generation metabolic peptide that activates three different hormone receptors at once (GLP-1, GIP, and glucagon) — the same ones the body uses to regulate appetite, blood sugar, and fat metabolism. Research suggests Eli Lilly’s 2023 Phase 2 trial produced the largest weight-loss effect recorded for a peptide to date.

Mitochondrial Peptides
MOTS-c
A short peptide that the mitochondria — the cell’s energy-producing structures — make themselves. Researchers study MOTS-c to understand how it signals to the rest of the body about energy balance. Research has examined how MOTS-c may affect glucose utilisation and fat metabolism in research models.

Why This Combination
Retatrutide acts mostly at the top of the system. Research suggests that by activating three hormone receptors (GLP-1, GIP, and glucagon), it acts on appetite signalling, insulin sensitivity, and energy expenditure. MOTS-c works at the other end — inside mitochondria — through AMPK, a cellular energy sensor studied in fat and glucose metabolism.
Pairing the two lets researchers study what happens when intake signalling (retatrutide) and cellular energy use (MOTS-c) are examined together. One addresses supply, the other throughput.
Research Notes
Most published retatrutide research comes from Eli Lilly’s 2022–2024 trial program. MOTS-c has a longer academic history starting with Lee and colleagues at USC in 2015, with continuing work on its exercise-mimetic effects and its decline with age. Neither peptide is approved for therapeutic use in Australia.
How Researchers Use This Collection
Research designs typically include comparison groups — one peptide alone, the other alone, and both together — so researchers can see whether combining them adds up cleanly or produces stronger effects than expected. Common readouts include fasting glucose, HbA1c (a longer-term measure of blood sugar), and lipid profiles. Academic studies sometimes add markers of mitochondrial health.
